Dosage Information

Erectile Dysfunction (On-Demand Use)

• Recommended dose: 60 mg taken orally once, at least 30–60 minutes before sexual activity. This dose is not intended for daily use at this strength.
• Maximum limit: 60 mg in 24 hours (do not take more than once daily).
• Missed dose instructions: Not applicable for on-demand use. Take a dose only before planned sexual activity.
• Overdose instructions: Immediate medical attention is required. Common symptoms after overdosing may include severe headache, dizziness, fainting, chest pain, or prolonged erection (priapism).

Erectile Dysfunction (Daily Therapy – Off-Label for 60 mg Strength)

• Recommended dose: 60 mg tadalafil dosage is NOT recommended for daily therapy. Standard daily tadalafil dosing is lower (2.5–5 mg).
• Maximum limit: 60 mg in 24 hours.
• Missed dose instructions: If a dose is missed, skip the missed dose and take the next dose at its scheduled time. Do not double the dose.
• Overdose instructions: Immediately seek medical evaluation. Monitor for hypotension and priapism.

Benign Prostatic Hyperplasia (BPH) – Off-Label Indication

• Recommended dose: 60 mg is higher than standard dosing. The usual tadalafil dose for BPH is 5 mg once in 24 hours. 60 mg is generally not recommended.
• Maximum limit: Do not exceed 60 mg in 24 hours.
• Missed dose instructions: Take the missed dose as soon as remembering about it, unless the timing is close to the next dose. Do not double-dose.
• Overdose instructions: Immediately seek emergency care. Monitoring of blood pressure and cardiovascular status is required after an overdose.

Pulmonary Arterial Hypertension (PAH) – Off-Label Indication

• Maximum limit: Do not exceed 60 mg dose in 24 hours

How Tadarise PRO 60mg Works

Tadarise PRO 60 mg consists of 60 milligrams of the drug tadalafil. This drug focuses on the NO-cGMP pathway, which is crucial for the circulatory response to external stimuli. Any disruptions in this system result in conditions like ED and PAH.

Nitric oxide (NO) is typically released by nerve cells and endothelial tissues in reaction to external stimuli (sexual stimulation, endothelial shear stress). NO triggers the release of an enzyme, guanylate cyclase, into the surrounding tissues. This enzyme converts inactive guanosine triphosphate (GTP) into active cyclic guanosine monophosphate (cGMP).

cGMPs are essential secondary chemical messengers that control physiological activities in endothelial tissues. It reduces intracellular calcium levels and facilitates the entry of potassium ions into the cells. Consequently, cGMPs promote smooth muscle tissue relaxation and blood vessel dilatation (vasodilation).

In response to excess cGMP, another enzyme known as phosphodiesterase-5 (PDE-5) is released via a feedback mechanism. Interactions

1.Blood pressure-affecting agents

Certain agents lower the blood pressure due to several mechanisms. As Tadarise PRO 60 mg works by dilating the blood vessels, there is a hypotensive effect on the blood vessels. Examples of such agents include nitrates, nitric oxide donors, antihypertensive drugs, and alpha blockers.

2.CYP3A4- affecting drugs

CYP3A4 is an enzyme involved in the metabolism of tadalafil. Tadalafil stays for a longer time in the body when CYP3A4 inhibitors are used. This raises the chances of adverse events like priapism, hypotension, vision problems, etc.

Similarly, CYP3A4 inducers shorten the half-life of tadalafil by accelerating its metabolism. Because of the low plasma concentrations, Cenforce 100 mg does not have any physiological effects.

Additional PDE-5 inhibitors

Excessive vasodilation occurs when Cenforce 100 mg is used with other PDE-5 inhibitors. This can result in potentially fatal complications as well as excessively low blood pressure.
