1.Erectile Dysfunction (ED)

ED refers to the inability of an adult male to achieve or sustain an erection for a minimum required amount of time. Tadalis-SX 80 mg increases the blood flow to the penis and facilitates relaxation of penile erectile tissue. This medication is to be taken on an on-demand basis and not to be consumed more than once in 24 hours. The effects of Tadalis-SX 80 mg can last for up to 36 hours after consumption.

2.Premature Ejaculation (PE)

PE is caused by nervous system impairment, specifically, the serotonin-based pathways. As a result, the ejeculatory process occurs prematurely without any voluntary control. For this condition, Tadalis-SX 80 mg (specifically the Dapoxetine component) supports the serotonin activity in the brain. As a result, the ejaculatory reflex is delayed. This medication also improves control over ejaculation for a sufficient amount of time.

Dosage Information

Erectile Dysfunction (On-Demand)

• Recommended dose: (Tadalafil 10–20 mg) 1 tablet of Tadalis-SX 80 mg taken 30–60 minutes before sexual activity (in this combination: 20 mg).
• Maximum limit: Tadalafil 20 mg per 24 hours.
• Other clinical information: Initial ED therapy is often 10 mg. The dosing may be increased to 20 mg based on effectiveness and tolerability. This dosing should not be taken more than once daily. It is not intended for continuous daily ED dosing when using this fixed combination.

Premature Ejaculation (On-Demand)

• Recommended dose: Dapoxetine 30 mg taken 1–3 hours before sexual activity (in this combination: 60 mg).
• Maximum limit: Dapoxetine 60 mg per 24 hours.
• Other clinical information: 30 mg of Dapoxetine is the usual starting dose. It may be increased to 60 mg if a 30 mg dose is ineffective and well-tolerated. No more than 1 dose of Tadalis-SX 80 mg must be consumed in 24 hours.

Concomitant Erectile Dysfunction + Premature Ejaculation

• Recommended dose: Tadalafil 20 mg + Dapoxetine 60 mg taken 1–3 hours before sexual activity (fixed-dose tablet).
• Maximum limit: Tadalafil 20 mg + Dapoxetine 60 mg per 24 hours.
• Other clinical information: The tolerability of Tadalis-XS 80 mg must be evaluated on an individual basis. This medication must not be combined with other PDE-5 inhibitors and SSRI drugs.

How Tadalis SX 80mg Works

Tadalis-SX 80 mg is a combination of two drugs- 20 mg Tadalafil and 60 mg Dapoxetine. These drugs target two different systems regarding male sexual function. Any impairment in these systems can lead to disorders like ED and PE.

Tadalafil in Tadalis-SX 80 mg is used in the treatment of ED. Under normal circumstances, sexual stimulation results in the release of a chemical known as Nitric Oxide (NO) from the nerve endings. The NO diffuses rapidly in the surrounding tissues and activates an enzyme called guanylate cyclase. This enzyme is responsible for converting guanosine triphosphate (GTP) into cyclic guanosine monophosphate (cGMP).

The cGMPs are key secondary chemical messengers that bring about the necessary physiological reactions in the corpus cavernosum (penile erectile tissue). It lowers the intracellular calcium and facilitates the entry of potassium ions into the cells. This leads to the relaxation of smooth muscle tissues. cGMP molecules also lead to dilation of blood vessels (vasodilation). These procedures lead to erection formation.

After a certain time, another enzyme called phosphodiesterase-5 (PDE-5) is released as a feedback mechanism to excess cGMP. It breaks down the cGMP molecules, which fades the effects of the cGMP molecules. As a result, the penis reverts to its resting state.

This reaction is essential to restore the normal functioning of the body. However, in some individuals, the effects of PDE-5 could be excessive or premature. This is the foundational mechanism that gives rise to ED.

The Tadalafil in Tadalis-SX 80 mg selectively targets the PDE-5 enzyme. It allows the cGMP molecules to follow through their physiological effects. 

The next component of Tadalis-SX 80 mg, Dapoxetine, is used in the treatment of PE. The ejaculatory process is normally controlled by the serotonin (5-HT) pathways in the brain and spinal cord. Serotonin is an inhibitory signal that is constitutively present on the synapses of the nerves.

In the presence of sexual stimulation, serotonin competes with other excitatory (pro-ejaculation) signals. When the concentration of the excitatory signals exceeds that of serotonin, the nervous system propels the body towards ejaculation. There are serotonin transporters (SERT) present in the synapses, which rapidly bind to serotonin and transports them back to the parental nerve cell. This interrupts the inhibitory signal for ejaculation. 

In some individuals, due to several underlying conditions, the serotonin uptake in the nerve cells is premature or excessive. This interruption of the inhibitory signal leads to premature ejaculation.

The Dapoxetine component of Tadalis-SX 80 mg selectively targets SERTs, thus allowing serotonin to persist in the nerve synapses for a sufficient amount of time. As a result, the ejaculatory process is activated after a longer time.
